NG2 cells as receivers and transducers of neuronal network signals via locally translated mRNA

Cells expressing the proteoglycan NG2 are precursors (OPC) to myelinating oligodendrocytes in development and disease. Stimulation of neurons synapsing with NG2 glia leads to activation of glutamate (AMPA) and GABAA neurotransmitter receptors on the NG2 cells. NG2 glia can be modulated e.g. by altering their synaptic input. The aim of the project is to characterise already identified process- and synapse-localised mRNAs in NG2 cells and the regulation of their translation in response to neuronal network activity. We will focus especially on Olig1 and Olig2, for whom we have already a large amount of data. In both these examples, the proteins play a role in the cell nucleus. Our working hypothesis is that the signal-induced local translation that we aim to unravel couples nuclear functions of these proteins to neuronal network input. This would entail that the newly synthesised proteins traffic back to the nucleus. Our results will hopefully contribute to a better understanding of regulation of the differentiation of NG2 cells to myelinating oligodendrocytes, as well as possibly help elucidate additional functions that these cells perform in the neuronal network. » weiterlesen» einklappen
