Aggregated proteins as associated with neurodegenerative disorders have been shown to be primary targets of selective autophagy involving the co-chaperone BAG3. In this project this process will be investigated focusing on substrate recognition and requirements, kinetics of aggresome targeting, and its regulation by endo- and exogenous factors. The role of post-translational modifications for BAG3 function will be analyzed as well. To get a detailed insight into the dynamics of the autophagy network, the BAG3 interactome will be characterized under different cellular conditions and the crosstalk with other autophagy routes (e.g. mitophagy) will be assessed.
