Streptococcus pneumoniae is a globally distributed pathogen that colonizes the nasopharynx and can cause diseases of the upper and lower respiratory tract. Until now, such infections could be effectively controlled by the use of antibiotics. However, due to the increasing development of resistance, clones are developing that no longer respond to such therapy. In order to find new therapies and treatment options against resistant pneumococcal pathogens, the transmission and colonization characteristics of the resistant clones must be analyzed on the one hand, and the mode of action of the host immune system must be understood on the other. New mechanistic insights will be used to develop potential strategies to influence the transmission success of S. pneumoniae and prevent progression to invasive lung disease. The focus of this work was to identify the genetic features that cause efficient spread and increased virulence of S. pneumoniae clones.
