This study aims to utilize TiO2, SiO2, gold colloids and organosiloxane NPs to examine their effects on lung surfactant form and function, particle interactions with a human alveolar-endothelial cell model and cell signaling, redox pathways, cell devision and other metabolic processes. Here the use of a primary human cell model of the alveolar blood -gas barrier is particularly relevant. The reactivity of NPs with the surfactant-epithelial-endothelial interface of the alveolar region is investigated. The studies will indicate differences in cellular responses relative to particle reactivity, uptake and translocation which will be relevant to mechanisms of action in vivo.
